AZD1446 is a selective modulator of the alpha4beta2 neuronal nicotinic receptor which arose from collaborative research conducted by both Targacept and AstraZeneca. The two companies signed terms under a 2005 collaborative research and license agreement with AstraZeneca being responsible for funding the development and potential commercialisation of AZD1446.

It is expected that AZD1446 will undergo a phase 2 study as an adjunct treatment of donepezil in patients with mild to moderate Alzheimer's disease. The decision by AstraZeneca to invest further in developing AZD1446 is testament to the continued promise of the alpha4beta2 NNR mechanism in Alzheimer's disease.

Donald deBethizy, the President and CEO of Targacept has stated that the planned evaluation of AZD1446 as an augmented treatment in Alzheimer's disease compliments Targacept's Phase 2b study of AZD3480 as a monotherapy as both Targacept and AstraZeneca search for a successful treatment for this major unmet medical need.

Alzheimer's is a degenerative disorder which progressively attacks the brain's nerve cells or neurons which results in the lost capacity to memorise, think and use language to converse.
